Abstract

Alzheimer’s disease (AD) is characterized not only by cognitive decline and memory impairment but also cause significant disturbance in sleep and circadian rhythms. Sleep disturbance is increasingly recognized as both a symptom and risk factor to AD progression. Amyloid-β (Aβ) accumulation has been implicated in neuronal dysfunction and altered sleep-wake regulation. The locus coeruleus (LC), the primary source of norepinephrine in the brain, plays acritical role in arousal and sleep-wake control and may be affected earlier by pathological changes associated with AD. This study aimed to characterize sleep architecture and homeostatic sleep responses in APP-SAA-KI mice, a knock-in mouse model of AD which develops progressive Aβ pathology.
